Pharmacokinetic properties of Amoxicillin
- Amoxicillin is resistant to gastric juice after oral intake and quicly absorbed in the gastrointestinal tract.
- Amoxicillin easily penetrates into the soft tissues.
- In 60 minutes after oral intake, Amoxicillin concentration reaches the peak.
- Amoxicillin concentration in plasma and interstitial fluid is maintained for 6-8 hours after oral intake.
- About 60% of Amoxicillin is eliminated unchanged by the kidneys.
Amoxicillin Indications
Amoxicillin is indicated for prophylaxis and treatment:
- ENT infections, caused by: α- and β-hemolytic strains Streptococcus spp; S. Pneumoniae; H. Influenzae or Staphylococcus spp. (recurrent tonsillitis, otitis media, sinusitis).
- Infectious diseases of the urogenital system, caused by: E. coli; E. Faecalis or P. mirabilis (pyelonephritis, cholecystitis, cystitis, pyelitis, cervicitis, cholangitis).
- Infectious skin diseases caused by: α- and β-hemolytic strains Streptococcus spp; Staphylococcus spp. or E. Coli (erysipelas, secondarily infected dermatitis, impetigo).
- Infectious diseases of the lower respiratory tracts caused by: α- and β-hemolytic strains Streptococcus spp; S. Influenzae; Staphylococcus spp. or S. Pneumoniae (chronic bronchitis, acute bronchitis, lobar pneumonia, bronchopneumonia)
- Ano-genital infection caused by N. Gonorrhoeae (gonorrhea)
For prophylaxis of duodenal ulcer infected with gram-negative bacterium, H.pylori, Amoxicillin is indicated in combination with Clarithromycin and proton pump inhibitor Lansoprazole.
Amoxicillin Contraindications
The only contraindication to Amoxicillin is an allergic reaction to Amoxicillin or hypersensitivity to penicillin antibiotics, such as: Penicillin; Oxacillin; Ampicillin; Carbenicillin; Ticarcillin; Azlocillin; Piperacillin.
Amoxicillin and pregnancy
- Amoxicillin is indicated during pregnancy only if a bacterial infection may harm fetus.
- Like any antibiotic, Amoxicillin reduces effectiveness of combined oral contraceptives. Women with childbearing ability taking estrogen progesterone contraceptives during treatment with Amoxicillin are recommended to use alternative methods of contraception (spermicides; condoms).
Amoxicillin Side Effects
- Nausea, vomiting or diarrhea can be observed first 2-3 days of Amoxicillin use.
- The most common side effects of Amoxicillin are observed in patients with allergy or hypersensitivity to penicillin antibiotics and in patients with hay fever, hives or asthma.
- A risk of mucosal candidiasis, hemorrhagic colitis or pseudomembranous increases at long-term Amoxicillin use.
- In children, penicillin antibiotics (including Amoxicillin) may cause the teeth discoloration. This side effect of Amoxicillin rarely occurs, and in regular tooth brushing is reversible.
